Can I accept a ‘Buy Now’ offer made via a question?

Yes you are correct, you cannot add a buy now once bids have been made.

You could (could ...) - manually remove all the bids, and then add a buy now ... But honestly dont do that, once you start doing these things, it can make the whole trade messy. Just dont.

Say to your bidder, " I am going to let the auction run, and you never know! Your winning bid, maybe less than a buy now anyway, you never know your luck!"

If you are Happy with the Offer you can change the Buy Now to the agreeable price unless you have other bidders & the Reserve has been met - if the Reserve is met you can remove bids back to under the Reserve price then change the Buy Now. - If it is a Buy Now only Listing you can change the price as long as it isn't a multi quantity listing & purchases have been made.

Most important - don't make any changes until you know your Buyer is online ready to purchase otherwise someone else may swoop in before them - Cheers.
